In September 2018, Hugo Vicente Miranda became a Principal Investigator and Affiliated Assistant Professor at Universidade NOVA de Lisboa, NOVA Medical School, in Portugal. Protein misfolding is a central problem in biology and is accepted as the major underpinning of neurodegenerative diseases such as Parkinson's, Alzheimer's and Huntington's disease. Vicente Miranda research has been focused on identifying strategies that prevent or halt the neurotoxicity induced by misfolded proteins, in particular of alpha-synuclein (aSyn). There is a strong association between diabetes and neurodegenerative diseases. Notably, young type-II diabetes patients have 380% higher risk of developing PD. Moreover, diabetes elicits faster motor decline progression, cognitive decline and lower binding of dopamine to its transporter in the striatum. During his PhD and Post-doc training, Vicente Miranda established that glycation is an underlying link between diabetes and Parkinson's disease. He identified aSyn as a target of MGO glycation, triggering its pathology. At the Lab of Professor Tiago F Outeiro, a world-class leader in PD, he also discovered the mechanism underlying the potential of Sirtuin 2 inhibition as a therapeutic strategy for PD. In his first independent project, he identified that aSyn post-translational modifications are biomarkers of PD with high diagnostic potential. In 2018, Dr. Vicente Miranda established his own group, the Dysmetabolism in Brain Diseases (DysBrainD) laboratory at NOVA Medical School, Lisbon, Portugal. The major aim of DysBrainD is to unveil novel molecular targets, whose modulation, based in the repositioning of approved drugs, presents therapeutic utility. DysBrainD aims to validate their usage in models of the disease and perform pre-clinical validation research in cohorts of PD and diabetic individuals. Recently, his group determined the protective potential of Hsp27 in Parkinson's disease. He and his team also reported that Brain glycation triggers Parkinson's disease-like behaviour and neuropathological features, by exacerbating glutamatergic signaling and by altering oxidative phosphorylation components. Dr. Vicente Miranda has published 29 papers in specialised journals (several manuscripts in top journals such as Brain, PLoS Biology, Nature Neuroscience), was awarded several grants and prizes, was invited for oral talks in several congresses, and participates in the peer review of papers and projects. Dr. Vicente Miranda won the Great Prize from Portuguese Diabetes Society in 2021, and is currently elected member of NOVA Medical School Council.
