My current scientific interest concerns the role of TrkB fragments on Alzheimer’s Disease (AD) pathophysiology. At the moment, I am finishing my Master thesis in Neurosciences at University of Lisbon about an intracellular fragment (TrkB-ICD) formed on this pathology. Indeed, Aß peptide accumulation leads to a sustained calpain activation, which promotes the release of TrkB-ICD fragment, due to calpain cleavage of TrkB-FL receptor, contributing to an unbalance between TrkB-FL receptor and truncated isoforms. Knowing that BDNF is specifically recognized by TrkB-FL receptor, its cleavage seriously impairs BDNF signalling and commits its functions involved in neuronal growth, differentiation and survival. Moreover, TrkB-ICD fragment is a stable protein, which after its release, is translocated to nucleus, where promotes a severe phosphorylation on several proteins on nucleus and also cytosol. However, it would be important to better characterize all the processes where this fragment can have a role and to explore the possibility of TrkB-ICD as an AD biomarker. I believe that articulation between my theoretical knowledge (Biochemistry and Neurosciences) and practical capabilities is important to ensure a successful academic career and helps to achieve the objectives proposed on this work, hopefully.
