Helena Florindo was awarded the Habilitation title in Pharmaceutical Sciences in 2018 by the University of Lisbon. She graduated in Pharmaceutical Sciences in 2003 and concluded her Ph.D. degree in 2008 in Pharmaceutical Sciences by the University of Lisbon, in collaboration with the University of London. Currently, she is a Full Professor in the Department of Pharmaceutics and Pharmaceutical Technology at the Faculty of Pharmacy, University of Lisbon. Since 2015, she has been the head of the BioNanoSciences ¿ Drug Delivery & Immunoengineering Research Group, at the Research Institute for Medicines (iMed.ULisboa), University of Lisbon. She has been actively involved in the training of a multidisciplinary team of 17 researchers, including 4 PostDoctoral fellows, 10 Ph.D. students, 4 master students, and two trainees, with a background in Nanotechnology, Immunology, Biotechnology, Biochemistry, Polymer Sciences, Cancer Biology and Chemical Biology. Both nanotechnology and immune-oncology fields have motivated her research, which has been focused on the rational development of novel chemically defined functionalized nanobiomaterials as novel immunotherapies for cancer treatment. These nano-immunotherapies are designed to overcome multiple tumor immunosuppressive properties by attaining specific immune cell targeting in pharmacological active sites, namely tumor microenvironment (TME) and metastasis. It includes the study of the transport mechanisms and effect at the molecular level of those novel nanomedicines on dendritic cell maturation and T cell activation, the characterization of their biodistribution profiles, and the evaluation of the in vivo efficacy and underlying impact on immune cell function, as adjuvants to improve the induction of an effector and specific anti-tumor immune response. Her major topics of research are: 1) Regulation of immunity by targeting dendritic cells (DCs) using nanotechnology-based tools to combine the antigen carry capacity of nanoparticles (NP) and the specific targeting and maturation of DC receptors in vivo. It aims to i) enhance antigen delivery to DC; ii) modulate antigen intracellular processing and presentation pathways; iii) block signaling pathways related to tumor evasion mechanisms; 2) Size-based targeting of lymph node-resident immune cells by nanoparticles to overcome major barriers for vaccine components and immunotherapies. 3) Characterization of bio-responsive materials impact on germinal center response to generally improve vaccine efficacy. 4) Dissecting tumor stromal and immune cell interplay to guide the design of multi-targeting nano-conjugates as innovative immunotherapeutic treatments against metastatic cancer diseases.
