Dr. Carvalho, PI at the Center for Neuroscience and Cell Biology (CNC), University of Coimbra (UC). She studies cellular and molecular mechanisms underlying diabetes, obesity and related complications. She has discovered that low insulin receptor substrate-1 protein levels in fat are predictors of pre-diabetes. She investigates insulin resistance and metabolic dysfunction in adipocytes and how immune cells infiltrate the adipose tissue to cause local tissue inflammation. She is interested in the effect of functional foods in combating insulin resistance and inflammation. Moreover, she has studied wound healing and tissue regeneration in diabetes, with the goal of enhancing early diagnosis of subjects at risk for chronic complications. For instance, she is assessing the role of microRNAs and antimicrobial peptides wound healing and infection. More recently, she was awarded several projects to do work on Non Communicable Diseases (NCDs), particularly obesity, diabetes and complications in Guinea-Bissau, West Africa. One funded by the European Foundation of the study of Diabetes, and others, funded by the Portuguese Cooperation, Camoes Institute. She is evaluating the prevalence of type 2 and type 1 diabetes as well as gestational diabetes, in Guinea-Bissau. She is very interested in studying the impact of mobility and migration on NCDs, particularly diabetes. In addition, a good portion of these projects are dedicated to capacitating and implementing health literacy locally in Guinea-Bissau. Dr. Carvalho and her team are advocates of Health Literacy and Implementation science for the public. She has published ˜170 scientific publications (articles and and abstracts) (h-index=42; >5000 citations; scopus). She has mentored: Post-Docs (˜20); PhDs (˜16); Master (˜20), Erasmus/lab internships (˜30). She has obtained ˜30 travel fellowships from the European Foundation for the Study of Diabetes (EFSD) for young students. She has secured funding from: Europe (H2020-MSCA-ITN-2016 TREATMENT; EFSD; Diabetes UK); NIH (NIGMS-NIH P20GM109096; Pepper Older American Independence Centers; ABI); National (INFARMED; FCT; SPD; SPT), and the Institute Camoes. She is a board member: European Tissue Repair Society, senior editor: J of Endocrinology; J Mol Endocrinology, and reviewer for ˜40 journals and international funding agencies: MRC UK; Israel Sci F; FONDECYT/CONACYT; FWF Der Wissenschaftsfonds; Wellcome Trust; Diabetes UK; French ANR, Horizon Europe PathFinder, and more recently she has been appointed Committee Member for Innovation Fund Denmark, etc. Main research lines: 1 - Insulin resistance 2 - Adipose physiology 3 - Wound healing and tissue regeneration 4 - Oxidative capacity of circulating cells and tissues 5. Cyclosporine A and tacrolimus reduce the amount of GLUT4 at the cell-surface in human adipocytes: increased endocytosis as a potential mechanism for the diabetogenic effects of immunosuppressive agents. Context:Immunosuppressive agentes are associated with profound metabolic side effects including new-onset diabetes and dyslipidemia after organ transplantation. Objective: Toi nvestigated the effects of the cyclosporine A(CsA)or tacrolimus ong lucose uptake and insulin signalling in human adipocytes and their impact on the regulation of celular trafficking of the glucose transporter 4 (GLUT4). 2. Neurotensin-loaded collagen dressings reduce inflammation and improve wound healing in diabetic mice. Impaired wound healing is an important clinical problem in diabetes mellitus and results in failure to completely heal diabetic foot ulcers (DFUs), which may lead to lower extremity amputations. In the present study, collagen based dressings were prepared to be applied as support for the delivery of neurotensin (NT), a neuropeptide that acts as an inflammatory modulator in wound healing. 4. mTOR inhibition with rapamycin cause impaired insulin signalling and glucose uptake in human subcutaneous and omental adipocytes. Rapamycin is an immunosuppressive agent used after organ transplantation, but its molecular effects on glucose metabolism needs further evaluation. We explored rapamycin effects on glucose uptake and insulin signalling proteins in adipocytes obtained via subcutaneous (n = 62) and omental (n = 10) fat biopsies in human donors. At therapeutic concentration (0.01 lM) rapamycin reduced basal and insul. 2012. 5. Improved Survival, Vascular Differentiation and Wound Healing Potential of Stem Cells Co-Cultured with Endothelial Cells. In this study, we developed a methodology to improve the survival, vascular differentiation and regenerative potential of umbilical cord blood (UCB)-derived hematopoietic stem cells (CD34+ cells), by co-culturing the stem cells in a 3D fibrin gel with CD34+-derived endothelial cells (ECs). ECs differentiated from CD34+ cells appear to have superior angiogenic properties to fully differentiated ECs. 2011. Pedroso, Dora C.
