José Marques Andrade

José Marques Andrade works in the fields of Microbiology and Molecular Biology, focused on RNA Biology. He has a solid background in RNA biology and protein manipulation and extensive knowledge on RNA-binding proteins and different RNA substrates, from the more labile mRNA to the more stable non-coding RNA. Dr. Andrade is dedicated to different aspects of RNA metabolism and in particularly, to the study of non-coding RNAs and their regulators in different bacteria. The main topic of Dr. Andrade's research has been the RNA metabolism, finding novel players and expanding the functions of RNA-binding proteins and non-coding RNAs, including rRNA and sRNA. For that, he studied both the Gram-negative Escherichia coli and the Gram-positive pathogen Listeria monocytogenes. Currently his work follows 2 main lines of interest: 1. Ribosomal RNA metabolism and Ribosome biogenesis His team has identified a novel function for the RNA chaperone Hfq as ribosome assembly factor. This expanded the regulatory spectrum of this RNA-binding protein beyond the most widely known function in promoting sRNA-mRNA basepairing. Hfq was found to bind to immature 30S ribosomal subunits, enriched in precursor 17S rRNA in what may constitute a checkpoint for ribosome assembly. Furthermore, recently we found that Hfq is involved in a novel RNA Quality control mechanism, together with RNase R, for the elimination of defective rRNA fragments. Hfq-like enzyme exist in human cells and may constitute a novel therapeutic target for ribosome-associated diseases, known as ribosomopathies. 2. RNA-binding proteins and ncRNAs in infection Dr. Andrade is also interested in the study of two important RNA-binding proteins involved in pathogenicity: the exoribonuclease PNPase and the RNA chaperone Hfq. We have demonstrated that knockout mutants of each of these factors result in the strong attenuation of virulence in the intracellular bacterial pathogen Listeria monocytogenes. We aim to use these mutants to identify new targets involved in infection pathways. This level of knowledge can ultimately be used to develop new antimicrobial strategies to tackle bacterial pathogens. Since 2015, José Andrade was Principal Investigator of 3 FCT funded projects. With an h-index of 15, José Andrade has over 1100 citations and a total of 18 published articles in leading journals in the fields of RNA biology, Molecular Biology and Microbiology, including The EMBO Journal, mBio and npj Biofilms and Microbiomes. At present, he is first author in 8 publications and signs as corresponding author in 6 articles and 3 book chapters, has over 30 oral comunications and 50 posters in international and national meetings. He was Guest Editor for a Special issue on "Analysis of Bacterial Ribosomes and interacting factors" in the Journal "Microorganisms" (2021-2022) and reviewer of several journals like PNAS, RNA, JBC, Frontiers in Cellular and Infection Microbiology and Scientific Reports. José Andrade is committed to the advanced training of young scientists, and his supervisor activity includes the mentoring of 2 Postdoc, 6 Doctoral and 5 Master students, along several undergraduates. Dr Andrade and his team enjoy taking part in educational outreach programs, like the science events "ITQB Open Day", "Scientist goes to School", or the "Oeiras International Science Festival".
