Florence Janody was awarded my Ph.D on December 1999 by the Mediterranean University, before joining the laboratory of Jessica Treisman at the Skirball Institute in 2000 as a Postdoc. During her training, she investigated mechanisms by which signalling cascades govern distinct cell fate decisions through the interaction of their downstream effectors with the transcriptional machinery. This resulted in 9 research articles in the top 15% journals of the research area (5-year IF: 10,77), among which 5 as first author. During her postdoc, she identified genes encoding for cytoskeleton proteins in a mosaic genetic screen for novel co-factors of signalling pathways. This is with the goal of exploring the role of the cytoskeleton as an organizer of signalling activities that she established her independency. She joined the Gulbenkian Institute of Science (IGC, Portugal) in 2006, where she led the ACTIN DYNAMICS´s group and was awarded with 2 consecutive 5-year competitive contracts from the Portuguese Research Council (FCT) (Ciências 2007 and Investigator FCT-2013). Work from her lab indicated that the actin cytoskeleton has major impacts on cellular behaviours akin to those of cancer cells. She therefore restrained her focus on understanding how the cytoskeleton controls the activity of cancer signalling pathways. In 2018, she was invited to join i3S, as leader of the CYTOSKELETAL REGULATION & CANCER GROUP. She was awarded with a highly competitive 6-year Principal Investigator contract from FCT (CEEC 2022, starting in March 2023). Altogether, her lab contributed to unveil mechanisms by which the cytoskeleton governs the location, duration, and intensity of cancer signalling pathways activation through diverse mechanisms, ranging from force generation to the spatial distribution of signalling regulators in Drosophila epithelia and during breast cancer initiation and progression. As a GROUP LEADER, she gathered more than 900K for her research group. She trained 5 postdocs, 3 PhD students and 11 Msc students and published 21 senior author manuscripts (Average IF: 7.7), among which 15 as corresponding author (Average IF: 8,24), 9 in collaboration with researchers from national (Claudine Chaouiya; Jose Pereira Leal; Pedro T Monteiro, Raquel Seruca; Joana Paredes, Eurico Morais-de-Sá) and international institutions (Jochen Guck, Fernando Casares, Stein Aerts, Nicolas Tapon, Jessica White, Mark Peifer, Jessica Treisman). Altogether, she co-authored 31 publications in major multidisciplinary scientific journals with more than 1000 citations. 96% of my work has been published in Quartile Q1, her h-index is of 18 (Web of Science). Building up on their findings, the Janody´s lab is currently using multi-disciplinary approaches, combining experimental models (Drosophila melanogaster, inducible mammalian epithelial cell models, TNBC patient-derived organoids) and computational modelling to answer four specific questions: How cell shape, a direct readout of intrinsic forces exerted by the cytoskeleton on the cell membrane, constrains the signalling network to predispose for or to hamper tumour initiation? How cytoskeleton-adhesions scaffolds orchestrate the signalling network instructing benign lesions to progress to malignant cancer cells? How the cytoskeleton and forces by the extracellular matrix propel TNBC cell invasion? How cytoskeleton-adhesions scaffolds supervise chemoresistant signalling networks in TNBC? Her work is expected to shed light into the rationale behind cancer signalling network. This knowledge should guide awareness on mechanisms by which the transcriptional programme is supervised by cell architecture to control cell fate decision during normal and disease development. Importantly, it may ultimately translate into tools that are relevant for the therapeutic intervention of cancer.

  2. Actin stress fiber organization promotes cell stiffening and proliferation of pre-invasive breast cancer cells. Studies of the role of actin in tumour progression have highlighted its key contribution in cell softening associated with cell invasion. Here, using a human breast cell line with conditional Src induction, we demonstrate that cells undergo a stiffening state prior to acquiring malignant features.   3. Subunits of the Drosophila actin-capping protein heterodimer regulate each other at multiple levels. The actin-Capping Protein heterodimer, composed of the a and ß subunits, is a master F-actin regulator. In addition to its role in many cellular processes, Capping Protein acts as a main tumor suppressor module in Drosophila and in humans, in part, by restricting the activity of Yorkie/YAP/TAZ oncogenes.
