Dr. Philippe Pierre is an international leader in dendritic cell biology and innate immunity research. During his Ph.D at the European Molecular Biology Laboratory (EMBL, HD, Germany), his research focused on the microtubule-binding protein, CLIP-170. After his post-doctoral training at Yale University School of Medicine (USA), where he focused on MHC II restricted antigen presentation, he moved to France in 2000. He created his laboratory at the Centre d’Immunologie de Marseille-Luminy (CIML, CNRS, France), where he focuses on the cell biology of dendritic cell activation with projects studying proteolysis, membrane trafficking, stress pathways, and microbial detection. He is CNRS director of research since 2006 and was director of CIML from 2018-2023. Many of his discoveries have direct clinical applications in the treatment of cancer and inflammatory diseases, while his keen interest in protein aggregation and tRNA modifications has allowed him to be recruited as a team leader at the institute of BioMedecine (IBIMED) and adjunct professor at the University of Aveiro in 2014, through the support of an Ilidio Pinho Chair of molecular immunology. His team at IBIMED carries out research on stress and Interferon responses to various biological insults. Dr. Pierre has published over 110 articles in peer-reviewed journals. Dr. Pierre is also member of the EMBO YIP and FSER programs. Philippe Pierre and his team focus on gaining fundamental knowledge of the biology of dendritic cells (DC) in normal and pathogenic conditions. DCs have the unique capacity to initiate primary immune responses by stimulating naive T cells. The team is interested in the biochemical changes occurring during DC activation by microbial products and pathogens (MAMPS). The team carries-out the functional dissection of these various processes, providing new insights to how a complex immune response could be potentially modulated for therapeutic purposes. The laboratory has accumulated an extensive scientific know-how in the study of DCs, and repetitively proven that it is able to transform innovative concepts in coherent projects that are all integrated in one global experimental strategy. In the evaluated period, we have focused on the importance of protein synthesis regulation for antigen presentation and cytokines production, as well as the membrane traffic steps necessary for activation by microbes and the acquisition by DC of their unique immune-regulatory functions. Our expertise allowed us to explore uncharted research areas by bridging very different research specialties. This originality has been acknowledged by IBIMED SAB and the research community, who supported the publication of most of our original works in excellent to exceptional journals (JCR ranking), as well as several news and views published on our work (Nature Reviews Rheumatology 2018 doi:10.1038/nrrheum.2018.25). The team was first to demonstrate the importance of the small RNA miR-155 in controlling DCs activation (Ceppi et al. PNAS 2009). The observation that regulation of mRNA translation and induction of stress pathways are central to DC function, motivated its most recent work on the role of the integrated stress pathway in the control of type I IFN transcription and translation during microbial detection. The laboratory has also developed powerful technologies to monitor protein synthesis and energy metabolism using puromycilation and immunodetection techniques, like flow cytometry, that have considerably facilitated measurement of these activities at single cell level and are now commonly used in more than 1000 laboratories worldwide (Schmidt et al., Nature Methods 2009, Arguello et al, Cell Metabolism 2020).
