Qian Wu
Qian Wu graduated with a BSc in Biological Sciences from Fudan University, China. He was awarded a PhD in molecular biology
and biochemistry at the University of the Chinese Academy of Sciences. During 2009-2016 as PhD candidates in CAS and 3 years
as Post-doc(2016-2019) at Zhejiang University, he has been working on the intrinsic mechanism of iron metabolism regulation
and a novel function of an iron-regulatory gene---Hemojuvelin. In the past 9 years, his colleagues and he demonstrated the
physiological function of macrophage ferroportin in mice. He also participated in a functional study of Steap3(a reductase
for ferric iron-ferrous iron transition in late endosome) In the 3rd year as a PhD candidate, he started his own project:
distinguishing the regulatory role of Hfe and Hemojuvelin (Hjv) in hepcidin regulation pathway. Using Hfe KO mice, Hjv KO
mice and Hfe/Hjv double KO mice, I finally demonstrated that Hfe function in hepcidin regulation depends on the presence of
Hjv. Thus, Hfe turned out to be upstream of Hjv in regulating hepcidin. He also carried out a project about the novel anti-infection
function of Hjv. He found that Hjv KO mice were susceptive to bacterial peritoneal infection. Unexpectedly, iron/hepcidin
were not the only factors that cause this phenotype in Hjv KO mice. He eventually concluded that Hjv may function as a component
in Il12/Stat4 in regulating macrophages functions. In the meantime, He participated ferroptosis study in the Fudi lab. In
particular, his colleagues and himself demonstrated ferroptosis existence in hemochromatosis mice and indicated a pivotal
role of ferritin H in alleviating ferroptosis in iron-loaded mice. His other projects in Fudi lab included metal metabolism
genes function in hematopoiesis and host defense involving adult HSC differentiation block and hepatocyte-peripheral immune
system dialogue, respectively. Qian Wu joined Dr Miguel Soares laboratory in July 2019. He is now focusing on the iron metabolism
conferred disease tolerance in malaria,
Related Skills: During his PhD candidate and Postdoc period, he was well trained in mouse inbred, genotyping, establishing
pathological mice model, such as peritoneal infection model / CLP model/LPS-D-G sepsis model, and iron/Zinc enrich/deficient
mice model. Mouse model establishment is actually one of his interests. He also tried bone marrow transplant, adoptive transfer,
partial hepatectomy model, acute liver failure model(CCL4 and conA models). Correspondingly, He is good at i.p. injection
and i.v. injection(>90% successful injection for i.v. ). To further investigate the phenotype of specific cells in these mice
model. Primary cell isolation and culture is also an indispensable skill, he is familiar with primary hepatocytes/ peritoneal
macrophages/bone marrow-derived macrophages/splenocytes/islets isolation and further cell culture. He also learned intestinal
epithelium isolation in the Fudi laboratory. Common molecular biology, cell biology and biochemistry techniques are regular
experimental methods for him, such as plasmid construct, primer design, qPCR, western blot, ELISA, co-immunoprecipitation,
IHC/IF, flow cytometry. He also grasped plenty of methods/skills from his own project. For iron phenotyping, non-heme tissue
iron determination, serum iron/UIBC assay, heme content assay, Prussian blue staining, iron-DAB staining were included. For
the hemojuvelin project, he learned Nitric oxide determination, ROS determination, organ bacteria C.F.U. assay, macrophage
phagocytosis assay/killing assay.
